I would concur with DSD's comments...especially on the western end (Clinton-Sedalia/Boonville) of the trail...I run 28s with GREAT flat protection with good luck.
In addition to many, many day trips, have ridden Clinton-St. Charles several times (the last being in June, 2012) and have settled on 28s.
I know others like wider tires, that's just my 2 cents.
If it has been a really wet early March, I would consider non-knobby, kevlar 32s, but I can't imagine needing anything wider.
Again - that's just my 2 cents.
